Comparison of Wound - induced Proteins in Potato by Two - Dimensional Gel Electrophoresis


Abstract
Potato leaves were harvested at 0h, 6h, 12h, and 24h after wounding. SDS-PAGE showed that approximate molecular weights of 18- to 22-kDa proteins were newly synthesized in response to wounding. These proteins were detected at 6h and were still observed 24h after wounding. More than 100 proteins were resolved from two-dimensional gel electrophoresis. Twelve proteins from control leaves disappeared upon wounding, whereas over 20 proteins were newly synthesized in response to wounding. Immunoblot analysis showed that a 27-kDa protein from wounded leaves and non-wounded upper leaves from wounded plants reacted with gel-purified 22-kDa potato proteinase inhibitor specific antibody. The level of expression was low at 6h and was highest at 24h after wounding.
